Biological Activity

E0924G is a small molecule osteoprotegerin (OPG, EC50=0.29 uM) and runt-related transcription factor 2 (RUNX2) upregulator, effectively promotes osteogenesis and inhibits osteoclastogenesis through activating the PPARδ signaling pathway.
E0924G upregulates OPG expression in osteoprotegerin (OPG) with EC50 of 0.29 uM based on an established UOP cell model stably transfected with pGL4.17-OPGp.
E0924G significantly increases the protein levels of the osteoblast markers OPG, BMP2, and RUNX2 in a dose-dependent manner.
E0924G inhibits RANKL-induced osteoclast differentiation and bone resorption.
E0924G (5 mg/kg, 20 mg/kg, 0.5 % CMC-Na) attenuates bone loss in ovariectomized (OVX) rats, prevents bone loss in OVX rat model of osteoporosis.
E0924G attenuates bone loss in senescence-accelerated mouse prone 6 (SAMP6) mice, promotes bone formation and inhibits bone resorption.
E0924G promotes osteogenic differentiation and inhibits osteoclast differentiation through activating PPARδ.
