E567 is a small molecule TLR2 signaling inhibitor with anti-viral activity, efficiently inhibits both LCMV and HSV-1 induced cytokine responses in both human and mouse cell cultures.
E567 down-regulates TLR2 expression, and blocks LCMV-induced TLR2-mediated NF-κB activity and production of IL-8 in HEK cells, does not affect TNF-α-induced expression of TLR2.
E567 inhibits LCMV-induced TLR2 expression and LCMV replication in HEK293 cells.
E567 inhibits both LCMV and HSV induced cytokine responses in primary mouse macrophages.
E567 inhibits both LCMV and HSV-1-induced RANTES and IL8 production in mouse primary macrophages.
E567 (16 mg/kg) blocks LCMV and other TLR ligands induced inflammation, also significantly blocks both TLR2 ligand Pam2CSK4 and TLR4 ligand LPS induced IL-6 and MCP-1 production in vivo.
