ELOVL6 inhibitor 1w (ELOVL6-IN-4) is a potent, selective, and orally available ELOVL6 (long chain fatty acid elongase 6) inhibitor with IC50 of 79 and 94 nM for human and mouse ELOVL6 respectively. ELOVL6 inhibitor 1w shows high selectivity over the other ELOVL subtypes ELOVL1,2,5 (IC50>5 uM) and mouse ELOVL3 (IC50>10 uM). ELOVL6 inhibitor 1w effectively reduced the elongation index with IC50 of 30 nM in mouse hepatocyte cell H2.35 assay. ELOVL6 inhibitor 1w (1-10 mg/kg, p.o.) potently and dose-dependently suppressed the elongation index in the liver in mice.
