EPB-113 is a potent replication inhibitor of HCoV-229E with EC50 of 0.48 uM, also suppresses SARS-CoV-2, binds to hexameric nsp15 protein.
EPB-113 shows an EC90 value of 0.65 µM based on RT-qPCR for viral genome copies and of 0.40 µM based on titration of infectious virus inHCoV-229E-infected HEL299 cells.
EPB-113 inhibits SARS-CoV-2 replication in A549ACE2/TMPRSS2 cells. The molecule was shown to have an EC90 value (by RT-qPCR for viral load) of 12 µM and to produce more than 5-log10 reduction in viral load at 50 µM.
The antiviral effect of EPB-113 is tightly linked with nsp15, and specifically its EndoU domain, enhances the EndoU cleavage reaction
EPB-113 is active in both IFN responsive and non-responsive cell types.
EPB-113 acts at a post-entry and intermediate stage in first-cycle HCoV-229E replication.
