Endosidin17 (ES17) is a small molecule targets retromer component VPS35, binds to VPS35 impairs interaction with RABG3f, causes dose-dependent stabilization of VPS35 with EC50 of 0.9 uM.
ES17 inhibited the vacuole accumulation of PIN2-GFP in the dark.
After ES17 treatment, PIN2-GFP accumulated in cytoplasmic structures that colocalized with the endocytic marker FM4-64 and the TGN/PVC marker ARA7-RFP in a concentration-dependent manner.
VPS35-RABG3f binding precedes the release of RABG3f–GTP to interact with the vacuolar homotypic fusion and vacuole protein sorting (HOPS) complex, which is required for fusion of late compartments with the tonoplast.
