Biological Activity
FLF-15 is a potent, selective ADAM17 inhibitor with IC50 of 10.43 nM, selectively inhibits its IL-6R cleavage activity, inhibits HCC migration and invasion in vitro and in vivo.
FLF-15 does not inhibit ADAM17 enzymatic activity.
FLF-15 effective with IC50 values for human HCC cell lines MHCC97H and LM3 nearly 7-fold lower than that for normal human hepatic stellate cell LX-2 (2.13 µM and 2.53 µM vs. 15.32 µM, respectively.
FLF-15 inhibits migration and invasion in HCC cells in vitro.
FLF-15 (25 and 50 mg/kg) suppresses the lung metastasis of liver cancer in mouse model.
FLF-15 selectively suppresses IL-6 trans-signaling pathways leads to inactivation of AKT and ERK.
