FOXA1/2 inhibitor compound T is a small molecule inhibitor of collaboration between FOXA1 and FOXA2, disrupts the FOXA1/2 function, binds to forkhead domain of FOXA1 of SPR KD of 100 uM.
Compound T also reduced the luciferase reporter assay driven by the homolog FOXA3.
Compound T also disabled the FOXA1 and FOXA2 forkhead domain from binding to an immobilized double-strand DNA oligonucleotide by biolayer interferometry (BLI).
Compound T disrupted the binding of all three nucleotide sequences (i.e., C, PAL, and 2C) to either the forkhead domains of FOXA1, FOXA2, and FOXA1/FOXA2.
Compound T reduced FOXA1 chromatin binding in LNCaP cells, reduced FOXA1 and FOXA2 chromatin occupancy in PC3 cells.
Compound T selectively suppressed the clonogenic growth of cell lines expressing FOXA1 or FOXA1/2.
