FRS2 ligand 7 is a small molecule ligand of the PTB domain of FGFR proximal adaptor protein FRS2, prevents FRS2 activation and interrupts FGFR signaling, inhibits invasion and tumor growth.
FRS2 ligand 7 inhibited basal and bFGF-induced phosphorylation of extracellular regulated kinase ERK (pT202/Y204) in DAOY, SW780, M059K, SKOV3, AGS, DMS114 and RT-112 cells.
FRS2 ligand 7 inhibited collagen I invasion in both bFGF-sensitive and bFGF-insensitive cell lines.
FRS2 ligand 7 acts on FRS2 and displays limited off-target activities.
FRS2 ligand 7 is comparable to that of the structurally and functionally unrelated FGFR inhibitor BGJ398.
FRS2 ligand 7 (200 mg/kg, once per day PO) blocked cancer growth in the SKOV03 model in vivo.
