Fascin inhibitor G2 is a small molecule inhibitor of Fascin (FSCN1), inhibits the actin-bundling function of fascin with IC50 of 5-8 uM, blocks tumour invasion and metastatic colonization.
G2 inhibited the actin-bundling activity of fascin, but has no inhibitory effect on actin polymerization itself and the actin-bundling activity of L-plastin.
G2 directly interact with fascin with Kd of 5-20 uM.
G2 inhibits fascin function in cells and impairs the filopodial formation.
G2 could inhibit breast tumour cell migration specifically, with no effect on the growth of MDA-MB-231 and 4T1 tumour cells.
G2 is a potent inhibitor of 4T1 tumour cell metastasis from the mammary gland to the lungs, and the actin-bundling activity as well as filopodia are critical for tumour metastasis in mouse models.
