SP11 is a potent inhibitor of activated Fis1 (mitochondrial fission protein 1), covalently binds specifically to Cys41 of activated Fis1, inhibits CPM fluorescence of Thr34Asp Fis1 with IC50 of 9.4 uM.
Fis1 mediates mitochondrial fission by recruiting large cytosolic GTPase, Drp1, to the mitochondrial outer membrane.
SP11 binding to Thr34Asp Fis1 is 1:1 and irreversible.
SP11 does not bind Cys41Ser Fis1 nor does it bind Fis1 Cys41Ser Val56Cys double mutant.
SP11 inhibited the dimerization of immobilized Thr34Asp Fis1.
SP11 (250 nM) reduces H2O2-induced mitochondrial ROS production and fragmentation and increased Drp1 translocation to the mitochondria in HK-2 cells in culture.
SP11 acts primarily by inhibiting Fis1-mediated Drp1 binding.
