GSK223 (GSK-223) is a specific small molecule inhibitor of the NOD1 signaling pathway, shows NOD1 inhibitory activity with IC50 of 1 uM in 293/hNOD1 cell assays, does not inhibit RIP2 kinase.
GSK223 does not inhibit any kinase against a panel of 300 kinases.
GSK223 inhibits Tri-DAP -stimulated IL-8 secretion (IC50=1 uM) in HCT116 colon carcinoma cells which endogenously express both NOD proteins at functional levels, with minimal or no inhibition of MDP-stimulated NOD2 signaling.
GSK223 (0.1-10 uM) antagonized the Tri-DAP induced increase in p-JNK levels and reduction in IκBα in 293/hNOD1 cells in a concentration-dependent manner, no effect on the same responses to MDP in 293/hNOD2 cells at >30 uM.
