HB-MIX1902 Supplement (10X) is an optimized serum-free supplement used to promote and maintain the growth, maturity and viability of iPS-derived hepatic and biliary cells. MIX1902 is provided as a 10X liquid and is intended to be used with Hepatocyte Culture Medium for optimal viability and prolonged survival of hepatocytes and cholangiocytes. Use:
• Use HB-MIX1902 Supplement to supplement Hepatocyte Culture Medium (e. g. HCM, HepatoZYME and Leibovitz L-15) for optimal viability and prolongedsurvival of hepatocytes and cholangiocytes.
• Use HB-MIX1902 Supplement to supplement William's E Medium for long-term survival of hepatocytes and cholangiocytes.
• HB-MIX1902 Supplement when used as a supplement to DMEM (High Glucose) is effective for theculturing of tumor lines of hepatic origin.
• Use HB-MIX1902 Supplement to supplement Kubota's Medium for expansion of hepatic and biliary progenitors /precursor cells.
Prepare complete Hepatocyte Culture Medium:
1. Thaw HB-MIX1902 Supplement (10X) overnight at 4°C.
2. Aseptically homogenize the HB-MIX1902 Supplement (10X) by pipetting, and then add 10% HB-MIX1902 Supplement (10X) (10 mL/dL) to the Hepatocyte Culture Medium before use.
Note: Remaining HB-MIX1902 Supplement (10X) may stored at −20°C to −5°C. Thaw aliquots as needed. Do not freeze-thaw HBMIX1902 Supplement (10X) more than twice. Once thawed do not leave thawed supplement at 4°C for more than two weeks.
3. Aseptically add 1% GlutaMAX™ Supplement to the Hepatocyte Culture Medium before use.
Note: For hepatic and biliary differentiation / maturation, the complete Hepatocyte Culture Medium may require additional supplementation with Oncostatin M (OSM), Hepatocyte Growth Factor (HGF) and Dexamethasone (DEX).
Once supplemented, the complete Hepatocyte Culture Medium is stable for up to two weeks when stored in the dark at 2-8°C.
