HEC96719 is a potent, selective tricyclic farnesoid X receptor (FXR) agonist with EC50 of 1.37 nM and 1.55 nM by TR-FRET and luciferase reporter assays, respectively. HEC96719 exhibits excellent potency superior to GW4064 and obeticholic acid in in vitro and in vivo assays of FXR activation. HEC96719 also shows higher FXR selectivity and more favorable tissue distribution dominantly in liver and intestine. HEC96719 significantly improves histologic NASH and fibrosis with lower efficacy dose than Obeticholic acid (OCA).
