HQY426 is a specific, orally bioavailable inhibitor of pre-messenger RNA endonuclease cleavage and polyadenylation specificity factor 3 (CPSF3) with binding IC50 of 0.32 uM, exhibits potent antitumor activity.
HQY426 suppresses cell proliferation and causes G2-M arrest and apoptosis in ovarian cancer cells.
HQY426 (10 mg/kg per day) exhibits potent therapeutic efficacy and low systemic toxicity in SKOV3 xenografts BALB/c nude mice.
HQY426 effectively exacerbated genomic instability by down-regulating DNA damage repair genes and thus acted in synergy with poly(adenosine 5'-diphosphate–ribose) polymerase inhibition.
