IPA (Inhibitor of Priming Activity) is a small-molecule compound that binds Sec18 (Kd=3.84 uM) and blocks SNARE activation, inhibits vacuole fusion at the SNARE priming stage with IC50 of 50 uM.
IPA blocks SNARE priming and competes for phosphatidic acid (PA) binding to Sec18.
IPA more potently and specifically inhibits NSF/Sec18 activity than does N-ethylmaleimide.
IPA is a helpful chemical tool to further elucidate SNARE-priming dynamics.
