Igf2bp1 inhibitor 7773 is a specific small molecule inhibitor of Igf2bp1, binds to RNA binding domains of Igf2bp1, inhibits Igf2bp1 binding of Kras 6 RNA with IC50 of 30 uM.
Igf2bp1 inhibitor 7773 shows no effect on a control RBP (La) binding to its target (Bcl2 RNA) or on a histone methyltransferease protein, G9a, catalysing methylation of a peptide.
Igf2bp1 inhibitor 7773 interacts with both RRM12 and KH34 with KD values of 1.5 and of 7.2 µM, respectively, but not the KH12, di-domains, in MST assays.
Igf2bp1 inhibitor 7773 inhibits in vitro Kras RNA binding of both Igf2bp1 and Igf2bp3 in MST assays, but not Igf2bp2.
Igf2bp1 inhibitor 7773 (20 uM) downregulates Kras and other Igf2bp1 target RNAs, Kras protein, and downstream signalling in H1299 cells.
Igf2bp1 inhibitor 7773 (5-20 uM) inhibits wound healing and growth in soft agar without affecting cell proliferation in H1299 cells.
