Biological Activity
JD03-02 is a potent, highly selective small-molecule inhibitor of TRPC1/5 heteromers with IC50 of 0.2 nM, exhibiting >10,000-fold selectivity for TRPC1/5 heteromers over TRPC5 homomers.
JD03-02 also selectively inhibits TRPC1/4 heteromers over TRPC4 homomers (IC50 = 6.7 nM vs. 2385 nM).
JD03-02 exhibits minimal activity against other closely related TRPC family members, including TRPC3, TRPC6, and TRPC7 (all IC50 >10 uM).
JD03-02 (10 mg/kg and 30 mg/kg) exhibits rapid and robust antidepressant and anxiolytic efficacy in rodent models, does not induce
hyperphagia or locomotor hyperactivity.
