JNJ-61393215 (Tebideutorexant) is a potent, selective orexin-1 receptor (OX1R) antagonist with pKi values of 8.17 and 8.13 for human OX1R and rat OX1R, respectively.
JNJ-61393215 displays >100 -fold binding affinity for human OX1R compared with human OX2R (pKi=6.12), also shows no significant affinity for a panel of other receptors, ion channels, transporters, and kinases (Eurofins).
JNJ-61393215 selectively promotes REM sleep in OX2R-knockout (KO) mice, attenuates CO2-induced fear and cardiovascular response in rats.
