KIO-100 (PP-001) is a novel potent, specific small molecule inhibitor of DHODH with IC50 of <4 nM, 150-fold higher potency than leflunomide (IC50 of 650 nM).
KIO-100 (PP-001) does not inhibit tyrosine kinase or cause hepatotoxicity like leflunomide since, like other small molecule DHODH inhibitors, KIO-100 (PP-001) does not need to be converted from a prodrug to its active form in the liver.
KIO-100 (PP-001) reduces the proliferation in a dose-dependent manner in human peripheral blood lymphocytes (PBL), also inhibits cytokine/chemokine secretion of human lymphocytes.
KIO-100 (PP-001) efficiently suppresses rat uveitis in a preventive as well as therapeutic mode.
