Biological Activity
KRAS4b inhibitor Compound 3 is a specific small molecule covalent modifier of canonical RAS isoform KRAS4b CaaX-box cysteine (C185), prevents its posttranslational processing and subsequent membrane attachment.
KRAS4b inhibitor Compound 3 (10 uM) decreased the level of membrane-bound GFP-KRAS4b G12D, with no or minimal effect on GFP-NRAS G12D in n HeLa cells and MEF cells expressing the G12V mutant of KRAS4b
KRAS4b inhibitor Compound 3 reduces KRAS4b G12V protein levels.
KRAS4b inhibitor Compound 3 blocks farnesylation and the subsequent posttranslational processing of KRAS4b in cells.
