Biological Activity
Keap1-Nrf2 inhibitor 24 is a high potent small molecule inhibitor of Keap1-Nrf2 with Ki of 10 nM, induces expression of NQO1 mRNA with EC50 of 110 nM in BEAS-2B cells with qPCR-based assays.
Keap1-Nrf2 inhibitor 24 markedly reduces the expression of NLRP3, a key pattern-recognition receptor (PRR) domain of the inflammasome in macrophage-derived THP-1 cells, prevents the cleavage of gasdermin D (GSDMD), a process otherwise mediated by active caspase-1.
Keap1-Nrf2 inhibitor 24 shows anti-inflammatory effects by downregulating NLRP3 inflammasome and STING signalling.
Keap1-Nrf2 inhibitor 24 activates specific Nrf2-controlled genes and key immune signalling pathways.
