Ki19003 (Ki 19003) is a potent, specific CCR3 antagonist, inhibits the CCL11-induced migration of CCR3-expressing L1.2 cells with IC50 of 20 nM.
Ki19003 does not inhibit CCL2- or CCL19-induced migration of CCR2- or CCR7-receptor-expressing cells even at 10 uM.
Ki19003 inhibits CCL11-induced migration of bone marrow–derived eosinophilic granulocytes in a concentration-dependent manner, shows no
cytotoxicity against the cells at 10 uM.
Ki19003 (3 or 10 mg/kg, oral) significantly inhibited the increase in the number of eosinophils in bronchoalveolar lavage fluid (BALF) of sensitized BALB/c mice.
Ki19003 also inhibited the increased production of transforming growth factor-beta1 in BALF and the amount of hydroxyproline in the lungs in a dose-dependent manner.
Ki19003 significantly attenuated allergen-induced subepithelial and peribronchial fibrosis.
