Biological Activity

LLQ-45 is a novel covalent agonist for p53 Y220C, restores the DNA-binding ability of p53 Y220C with EC50 of 24.97 uM, shows selective anti-proliferative activity against p53 Y220C.
LLQ-45 potently inhibits the proliferation of HUH-7 cells carrying the p53 Y220C mutation with IC50 of 1.35 uM, but not p53 WT-expressing cells.
LLQ-45 restores p53 transcriptional function at cellular level via key p53 target genes.
LLQ-45 forms covalent bond with Cys220 through Michael addition.
