MARPIN is a novel ATR-Chk1 pathway inhibitor that inhibits hydroxyurea (HU)-induced phosphorylation of Ser345 on Chk1 with IC50 of 7.7 uM.
MARPIN inhibits ATR-selective phosphorylation and sensitizes p53-deficient cancer cells to DNA-damaging agents.
MARPIN does not inhibit ATR catalytic activity in vitro, but acts in a mechanistically distinct manner to disable ATR-Chk1 function.
MARPIN shows synergistic effect with cisplatin in xenograft model.
