Biological Activity
MAT2A inhibitor ZS34 is a potent, selective, and orally bioavailable MAT2A inhibitor with IC50 of 13.7 nM.
ZS34 selectively suppresses the growth of MTAP-deficient cancer cells (HCT116 MTAP+, IC50=124 nM), but not HCT116 MTAP- cancel cells.
ZS34 demonstrates on-target engagement in MTAP-deficient cells, as evidenced by inhibition of SAM synthesis, reduction of SDMA levels, and induction of DNA damage.
ZS34 displays minimal hERG and UGT1A1 liabilities along with favorable oral pharmacokinetics.
ZS34 (10-50 mg/kg, po/qd) demonstrates significant antitumor efficacy without obvious toxicity in HCT116 MTAP–/– xenograft mouse model.
