Biological Activity
METTL9i is a first-in-class, highly potent, selective and cell-active methyltransferase METTL9 inhibitor with IC50 of 67 nM.
METTL9i exhibits high selectivity for METTL9 relative to other methyltransferases.
METTL9i binds within the S-adenosylmethionine (SAM) binding pocket of METTL9.
METTL9i engages METTL9 and leads to a reduction in global 1-methylhistidine (1-MH) levels in cells, without affecting 3-methylhistidine.
METTL9i displaysthe potent inhibitory activity against pancreatic cancer cells BxPC-3 and PANC-1 with IC50 values of 3.42 µM and 9.92 µM, respectively.
