Biological Activity

MIPO is a pharmacological small-molecule NEK9 inhibitor with SPR KD of 7.72 uM, inhibits viability of Huh-7 and Hep3b cells with IC50 of 18.8 uM and 19.7 uM.
MIPO reduces NEK9 mRNA and protein levels in Huh-7 and Hep3b cells, NEK9 overexpression partially rescued the inhibitory effects of MIPO, but not NEK2 or NEK6.
MIPO suppresses NEK9-driven tumour growth and restore antitumour immunity in vivo, restores CD8+ T cell antitumour activity.
Combining MIPO with PD-L1 blocker CA-170 synergistically enhances antitumour immunity in HCC.
