MLN120B dihydrochloride (ML120B) is a potent, specific and ATP competitive inhibitor of IKKbeta (IKKβ) with IC50 of 0.06 uM, blocks TNF-α-induced NF-κB activation in multiple myeloma cells through inhibition of IκBα phosphorylation and degradation of IκBα.
ML120B is greater than 1000 times more potent in inhibiting IKKβ than other inhibiting kinases, does not inhibit IKKα.
ML120B inhibited IL-6 and RANTES production in response to LPS in a dose-dependent manner with IC50 of 1 uM on LPS induced primary cells.
ML120B inhibits NF-κB activation and rapidly induces apoptosis in lymphoid organs in vivo.
