Biological Activity
MPI-13 is a potent, covalent, orally bioavailable P. falciparum 20S proteasome (Pf20S) inhibitor with IC50 of 12 nM, 230 nM and 24 nM for Pf20S β5, Hs20S β5c and Hs20S β5i respectively, exhibits fast-acting antimalarial activity with LD50 of 11 nM for Pf 3D7 strain.
MPI-13 inhibits male exflagellation in mature (stage V) gametocytes.
MPI-13 (25 mg/kg QDx4) is oral bioavailable and exhibits potent activity against Pf in vivo model of Pf malaria.
