Biological Activity

MS9024 is a potent, selective PROTAC degrader of DNA methyltransferase 1 (DNMT1) with DC50 of 40 nM in HCT116 cells, Dmax=89%.
MS9024 potently induces DNMT1 degradation in colorectal cancer (CRC), breast cancer (BC), and non-small-cell lung cancer (NSCLC) cells.
MS9024 selectively induced DNMT1 degradation in a concentration-, time-, and ubiquitin–proteasome system (UPS)-dependent manner without affecting DNMT1 transcription.
MS9024 potently degraded DNMT1 without altering the mRNA expression of DNMT1 in BC and NSCLC cells.
