Biological Activity
MT-228 is a potent, selective, first-in-class non-muscle myosin II (NMII) inhibitor with Ki of 1.5 uM and cytokinesis EC50 of 1.0 uM, 17-fold selectivity for NMII over cardiac myosin II (CMII).
MT-228 has improved cardiac safety and selectivity compared to Blebbistatin.
MT-228's brain penetrance was also significantly improved over Blebbistatin.
MT-228 is a four-fold more potent inhibitor of SmMII (KI = 4.0 µM) than CMII (KI = 17.0 µM).
MT-228 (0.5 mg/kg) produces a selective disruption of the motivation in METH-associated CPP in mice, without impacting other brain processes.
