Biological Activity

MTHFD1/2 inhibitor 34 is a potent, dual MTHFD1/2 inhibitor with IC50 of 0.26 uM and 0.031 uM for MTHFD1 and MTHFD2 respectively.
MTHFD1/2 inhibitor 34 exhibits dose-dependent inhibition of cell viability across five leukemia cell lines MOLM-13, THP-1, K562, NALM-6, and MOLT-4 cells with IC50 of 0.06 uM-1.78 uM.
MTHFD1/2 inhibitor 34 elicited pronounced, dose-dependent tumor regression, achieving a TGI of 79% at 300 mg/kg in MOLM-13 xenograft mouse model.
