Biological Activity
MTHFD2 inhibitor 31 is a potent, highly selective MTHFD2 inhibitor with IC50 of 38 nM, >180-fold selective over MTHFD1 (IC50=7440 nM).
MTHFD2 inhibitor 31 exhibits dose-dependent inhibition of cell viability across five leukemia cell lines MOLM-13, THP-1, K562, NALM-6, and MOLT-4 cells with IC50 of 0.25 uM-4.75 uM.
MTHFD2 inhibitor 31 induced modest, dose-dependent inhibition, with tumor growth inhibition of 37% at 100 mg/kg and 67% at 300 mg/kg in MOLM-13 xenograft mouse model.
