Motolimod (VTX-2337) is a potent, selective small-molecule TLR8 agonist, stimulates PBMCs to produce TNFα and IL-12 with EC50 of 140 and 120 nM, respectively.
VTX-2337 selectively activated TLR8, but not HEK293 cells transfected other TLRs (TLR2, 3, 4, 5, and 9), 30-fold over TLR7.
VTX-2337 drives mDCs and monocytes to produce IL-12 and TNFα via NF-κB activation, but not pDCs.
VTX-2337 stimulates the production of other immune mediators including IFNγ, activates NK cells to produce IFNγ.
VTX-2337 augments the lytic function of NK cells and enhances ADCC, enhances ADCC in donors with different SNP on FcγR3A.
VTX-2337 enhances rituximab-mediated ADCC, including individuals with FcγR3A genotypes associated with a reduced affinity for therapeutic monoclonal antibodies.
