Cardiac-myosin inhibitor F10 is a cardiac-myosin specific and allosteric inhibitor with IC50 of 23 uM on actomyosin ATPase activity.
F10 reduces the maximum rate of ATP hydrolysis, does not affect the affinity of myosin S1 for F-actin per se.
F10 (20 uM) significantly reduces the ATPase activity of fully Ca2+-activated cardiac myofibrils by about 25% but has no effect on the ATPase of myofibrils isolated from fast skeletal muscle, F10 is a MYH7/6-specific inhibitor.
F10 slows nucleotide release from bovine β-cardiac myosin. can stabilize both its biochemical and structural OFF state.
F10 reduces both isometric force and its calcium sensitivity in isolated muscle fibers and acts as a negative inotrope in Langendorff-perfused hearts.
