N1-hexyl-N5-benzyl-biguanide mesylate (HBB) is a potent and specific inhibitor of CYP3A4 AA epoxygenase activity with IC50 of 9.5 uM for inhibition of CYP3A4-mediated (±)-14,15-EET synthesis.
HBB displays significantly higher for CYP2C8 (IC50=45 uM), and does not inhibit the AA epoxygenase activity of CYP2J2.
HBB inhibits MCF-7 cell line and ER+ and triple-negative breast cancer cell lines with similar IC50 (20 uM).
HBB strongly inhibits the oxygen consumption rate (OCR) and suppresses ATP stores, rapidly activates pAMPK and inhibits Stat3, mTOR, and ERK.
HBB inhibits MCF-7 xenograft tumor growth (24 mg/kg/week i.p.).
