NEO214 (NEO-214) is a novel, BBB-permeable rolipram-perillyl alcohol conjugate, shows effectivity against both TMZ-sensitive and TMZ-resistant glioma cells (T98G IC50=40 uM).
NEO214 triggers apoptosis by activating the ER stress pathway, induces Death Receptor 5 (DR5) and TNF-related apoptosis inducing ligand (TRAIL/Apo2L) mediated cytotoxicity, without cytotoxicity to normal cells.
NEO214 is generated by covalently conjugating rolipram, a PDE4 (phosphodiesterase 4) inhibitor, to perillyl alcohol, a naturally occurring monoterpene related to limonene.
NEO214 decreases glioma progression in the in vivo mouse tumor model.
