NF764 is a potent and selective, covalent β-catenin (CTNNB1) binder and degrader wtih DC50 of 3.5 nM in HT29 cells (Emax=85%), targeting C619 of CTNNB1.
NF764 inhibits CTNNB1 transcriptional activity, demonstrating functional inhibition of the WNT pathway in cells.
NF764 treatment significantly reduced the levels of CTNNB1 target genes,, including MYC, S100A6, AXIN2, and CCND1.
FLAG-CTNNB1 C619S mutation expressing HEK293T cells are completely resistant to NF764-mediated CTNNB1 loss.
