Biological Activity

NGD-94-1 is a highly potent, selective dopamine D4 receptor antagonist with Ki of 3.6 nM.
NGD-94-1 displays 600-fold selectivity for the D4.2 receptor subtype compared with a wide variety of monoamine or other neurotransmitter receptor or modulatory sites except for 5-HT1A and 5-HT3 receptors.
NGD 94-1 completely reverses the decrease in forskolin-stimulated cAMP levels produced by the dopamine receptor full agonist quinpirole.
NGD-94-1 behaves as an agonist in human embryonic kidney 293 cells expressing human recombinant dopamine D4.4 receptors.
