Biological Activity
NGI-189 is a potent, small molecule inhibitor of oligosaccharyltransferase (OST) with IC50 of 0.09 uM.
Oligosaccharyltransferase (OST) is a ER membrane-embedded enzyme complex responsible for transferring glycans to newly synthesized proteins.
NGI-189 demonstrates activity against both OST-A and OST-B, with preferential inhibition of OST-A.
NGI-189 induces great accumulation in the G1 phase of the cell cycle in PC-9 cells, causes induction of the pro-apoptotic protein, Bim, and again without an increase in phosphorylation of other RTKs.
NGI-189 inhibits EGFR N-glycosylation, blocks EGFR dependent signaling in EGFR mutant NSCLC.
NGI-189 (10 mg/kg, i.p.) reduces tumor growth in mice bearing MDA-MB-231-ERLucT xenografts.
