Biological Activity

NN-01-195 is a chimeric small molecule conjugate of HSP90 and AURKA inhibitor, tightly binds and inhibits both AURKA and HSP90 with IC50 of 3.1 nM and 8.1 nM in biochemical assays.
NN-01-195 combines an AURKA inhibitor related to TAS-119/VIC-1911 with an HSP90-binding moiety related to SNX2112.
NN-01-195 causes mitotic arrest and spindle abnormalities and a profile of signaling changes that closely resembles that of an AURKA inhibitor.
NN-01-195 reduces cell viability and promotes mitotic abnormalities.
NN-01-195 elevates apoptosis and reduces cell viability for human papillomavirus–negative HNSCC (FaDu, Detroit 562, and Cal27), NSCLC (H1975, PC9), and melanoma (WM3000, WM33451) models with IC50 of 0.27-2.98 uM.
NN-01-195 (10 mg/kg, IP) is highly effective at controlling xenograft tumor volume in FaDu cells xenografted onto NSG mice.
