NSC126405 is a small-molecule DEPTOR inhibitor that prevent DEPTOR binding to mTOR, acutely activates mTORC1 and selectively induces MM cell apoptosis and cell cycle arrest, binds to recombinant DEPTOR but not mTOR.
NSC126405 is a small-molecule DEPTOR inhibitor that prevent DEPTOR binding to mTOR, acutely activates mTORC1 and selectively induces MM cell apoptosis and cell cycle arrest, binds to recombinant DEPTOR but not mTOR. NSC126405 prevents mTOR/DEPTOR binding within MM cell lines and SPR assays. NSC126405 cytotoxic effect against MM cell lines with the highest DEPTOR expression with IC50 of 0.8-2.5 uM. NSC126405 induces apoptosis and cell-cycle arrest in multiple myeloma cell lines and prevents outgrowth of human multiple myeloma cells in immunodeficient mice.
