NU6140 is a selective inhibitor of CDK2 with IC50 of 0.41 uM for CDK2-cyclin A, 10- to 36-fold selective for CDK2-cyclin A over the other CDKs. NU6140 inhibits cell proliferation and cell cycle progression and potentiates the apoptotic response to paclitaxel in HeLa cells in comparison with purvalanol A. NU6140 significantly increases the apoptotic rate (to a greater extent than that observed in the paclitaxel-purvalanol A combination), with inhibition of survivin expression/phosphorylation in combination with paclitaxel.
