NU7026 is a potent, selective inhibitor of DNA repair enzyme DNA-dependent protein kinase (DNA-PK) with IC50 of 0.23 uM. NU7026 is selective for DNA-PK over related phosphatidylinositol 3-kinase-related kinase (PIKK) family members. NU7026 (10 micro M) potentiated radiosensitization-induced DNA double strand break (DNA DSB) repair in DNA-PK proficient cancer cells. NU7026 potentiates the cytotoxicity of topoisomerase II poisons used in the treatment of leukemia.
