Biological Activity

Nedemelteon (Compound (S)-3b) is a poteny, orally bioavailable and BBB-permeable melatonin receptor (MT1 and MT2) agonist with binding Ki of 0.031 and 0.070 nM respectively.
Nedemelteon exhibits full agonism and superior MT1 and MT2 agonistic activities (MT1, EC50 = 0.015 nM; MT2, EC50 = 0.51 nM) using cAMP production in CHO cells expressing human MT1 or MT2 receptors.
Nedemelteon (10 mg/kg, p.o.) significantly increased the total sleep time in freely moving cats.
