Nicotinamide riboside (NR) is an orally active NAD+ precursor, increases NAD+ levels and activates SIRT1 and SIRT3, promotes peroxisome proliferator-activated receptor-γ coactivator 1 (PGC)-1α expression in the brain. Nicotinamide riboside supplementation in mammalian cells and mouse tissues increases NAD(+) levels and activates SIRT1 and SIRT3, culminating in enhanced oxidative metabolism and protection against high-fat diet-induced metabolic abnormalities. Nicotinamide riboside regulates β-secretase 1 degradation and mitochondrial gene expression in Alzheimer's mouse models.
