Biological Activity
OICR18704 is a potent, specific, allosteric inhibitor of human mitochondrial ClpP protease with IC50 of 13.7 nM and EC50 of 0.3 uM in cellular thermal shift assays (CETSA).
OICR18704 does not inhibit the ClpP homologue from Staphylococcus aureus and Neisseria meningitidis.
OICR18704 does not inhibit peptidase activity of human proteasome 20S, and clean selectivity against a panel of 468 human kinases at 1 uM.
OICR18704 destabilizes ClpP inter-ring interactions, impacts the conformational transitions of ClpP
OICR18704 (100 mg/kg, 5% DMSO + 47.5% Cremophor EL + 4.75% Tween-80, oral) reduced leukemic burden in xenograft OCI-AML2 AML model.
