OV329 (OV 329) is a highly potent inactivator of gamma-aminobutyric acid aminotransferase (GABA-AT) with Ki of 9.69 uM.
OV329 is 10 times more potent as an inactivator of GABA-AT than CPP-115, OV329 has favorable pharmacokinetic properties and low off-target activities.
OV329 (5-40 mg/kg, i.p.) dose-dependently increases the thresholds of intravenous pentylenetetrazole (ivPTZ)-induced seizures.
OV329 is dramatically superior to CPP-115 in suppressing the release of dopamine in the corpus striatum, which occurs subsequent to either an acute cocaine or nicotine challenge in vivo.
OV329 also attenuates increased metabolic demands (neuronal glucose metabolism) in the hippocampus, a brain region that encodes spatial information concerning the environment in which an animal receives a reinforcing or aversive drug.
