OXS007417 (OXS 007417) is a small moelcule that induces differentiation of AML cells in vitro (CD11b increasein HL-60 cell EC50=48 nM) and shows antitumor effects in vivo.
OXS007417 displays favorable pharmacokinetic properties and is well-tolerated at a therapeutic dose.
OXS007417 showed activity across multiple AML cell lines and displayed a mechanistic profile consistent with differentiation, caused a clear decrease in cell numbers in the panel of AML cell lines.
OXS007417 also caused morphological changes consistent with differentiation, such as a lighter cytoplasm, a higher cytoplasm-to-nucleus ratio, an increase in granulation, and a change in the cell shape and size.
At 10 mg/kg twice daily dosing, OXS007417 significantly delayed the growth of HL-60 tumors, with a T/C of 57%.
