Biological Activity
PGK1 activator L03 (AT-051/43421517) is a specific small molecule activator of phosphoglycerate kinase 1 (PGK1) with SPR KD of 2.45 uM, significantly enhances enzymatic activity of PGK1 at 0.25-2 uM in AβO-induced SH-SY5Y cells.
L03 (AT-051/43421517) (1-2 uM) significantly increases the PGK1 activity of SH-SY5Y cells not exposed to AβO.
L03 (0.25 and 1 µM) significantly protected SH-SY5Y cells from the AβO-induced impairment of cell viability, L03 (0.25 and 1 µM) itself has no significant effect on the viability of SH-SY5Y cells.
L03 ameliorates AβO-damaged SH-SY5Y cells by promoting glycolysis, reduced apoptosis, enhanced glycolytic metabolism, restored TCA cycle, and decreased ROS production in AβO-damaged SH-SY5Y cells.
L03 (0.5 and 1 mg/kg, i.p.) improves the learning and memory ability in 3×Tg-AD mice.
